One per sheet tiles a single label edge to edge, so every cut line runs the full length of the sheet and one cut serves two labels.

Mixed puts several labels on one sheet in horizontal bands, one label type per band. Different label heights can't butt against each other and still give straight cuts, so you slice the bands apart first, then cut each band into labels.

Before the first run on a new paper or shop, switch to Test print: it lays down one label and a 50mm bar. If the bar doesn't measure 50mm the machine is rescaling and the full sheet would be wrong by the same factor.
